چکیده

A mutant of Esckch ia coZi waa isolated which showed a requirement for methionine + lysine when a fairly large inoculum of washed organisms was used. Cystathionine or homocysteine replaced methionine, and diaminopimelic acid replaced lysine; no other amino acid or growth factor was active. The amounts of the amino acids required to promote growth were only about one-tenth of the quantity of methionine and lysine found in the grown organisms. A quantitative study with lV-glucose and 85S-sulphate confirmed that these amino acids were synthesized de novo. Although homoserine would not replace methionine for growth, the presence of unlabelled homoserine suppressed the incorporation of isotope from 14C-glucose into methionine and threonine, indicating that the accepted pathways are operative and that the block in methionine synthesis was in the formation of cystathionine. Small amounts of yeast extract replaced the amino acid requirement and decreased one-thousandfold the size of the inoculum required.

Isolation of a dihydrofolate reductase-deficient mutant of Escherichia coli.

A strain of Escherichia coli was isolated in which dihydrofolate reductase was not detected by an enzyme assay or by competition for antibody. This strain requires methionine, glycine, a purine, and thymidine for growth in addition to the auxotrophic requirements of the parent strain. It was found to be useful as a recipient of plasmids harboring dihydrofolate reductase genes.

Effect of canavanine on mutants of Neurospora and Bacillus subtilis.

The inhibition of microorganisms by canavanine, an amino acid whi -1 occurs in the jack and horse beans (1, 2), has recently been the subject of several investigations. Horowitz and Srb (3) studied the effect of canavanine on three wild type strains of Neurospora, one sensitive, one of intermediate sensitivity, and one resistant to canavanine inhibition. They found that arginine and, to a lesse...

Soluble Expression and Purification of Q59L Mutant L-asparaginase in the Presence of Chaperones in SHuffle™ T7 strain

Background and Aims: Q59L mutant of L-asparaginase enzyme from Escherichia coli (E. coli) has been introduced with lower side effects. This version of the enzyme might have potential applications in the treatment of leukemia patients. We utilized SHuffle T7 strain of E. coli, to produce the mutant enzyme in the presence of chaperone molecules. Materials and Methods: Q59LAsp gene was cloned in...
